實驗原理

CCK-8（Cell Counting kit-8）試劑中含有WST–8：化學(xué)名：2-(2-甲氧基-4-硝基苯基)-3-(4-硝基苯基)-5-(2,4-二磺酸苯)-2H-四唑單鈉鹽]， 它在電子載體1-甲氧基-5-甲基吩嗪硫酸二甲酯（1-Methoxy PMS）的作用下被細(xì)胞線粒體中的脫氫酶還原為具有高度水溶性的黃色甲臜產(chǎn)物（Formazan），生成的甲臜物的數(shù)量與活細(xì)胞的數(shù)量成正比，可采用酶聯(lián) 免疫檢測儀在450nm波長處測定其光吸收值，間接反映活細(xì)胞數(shù)量。
